Yes, you can use such a big HD, but the clusters would be HUGE, if you don´t split the drive up into a bunch of smaller partitions.
We just swapped IDE drive in the lab´s Falcon, from a Toshiba 2.5" 224MB, to a Quantum 3.5" 850MB. No problems, except the big clusters. (8152 bytes on a 212MB partition.)
If your new drive is a 2.5", then just plug it in. If it is a 3.5", you will need an adapter connector, cos the 3.5" flat ribbon cable is wider in size than the 2.5" cable in the Falc.
If you move the original 2.5" connector a bit down the cable, and place the new adapter connector up at the end, then you can use both drives at the same time. Just cut off leads 41-44, AFTER the 2.5" connector, cos the 3.5" drive only use 40 leads. Those last four only feeds the 2.5" drive´s motor with power.

IF you use KOBOLD to move the stuff from the old drive to the new one, then DON´T USE VERSION 3.5!! It´s buggy and WILL mess the new drive up completely! (happened to us) Version 3.0 is OK.
