Well, you already said it in your text:
The SIO2ST-cable connects to the SIO port on the XL-Side, just the port the original XL-Floppy would connect to as well.
Unfortunately, Atari ST diskdrives do not connect to the serial or ACSI-port but to the internal Shugart-Bus so going over ACSI, Serial or Parallel would necessarily involve software for the ST.
There might be a chance doing what you plan by removing the diskdrive from your ST and connect a cable directly to the Shugart-connector in the ST and wire that cable to the PC some way - preferably Parallel i think, but it would mean quite some effort on the (driver)software side on the PC.
